Summary

Invenra is seeking a highly motivated scientist/senior scientist with a background in immuno-oncology and immunology assay techniques. This individual will work as part of a multi-disciplinary team towards development of novel therapeutics using the Invenra platform technologies. Aspects of the job include in vitro and ex vivo immune cell-based functional assays (activation, differentiation, proliferation, suppression, target cell killing), cytokine analysis, primary cell isolation and differentiation and tissue culture techniques. Experience with in vivo models for immuno-oncology (syngeneic, humanized, GEMMs, etc.) is a plus. Knowledge, Skills and Abilities

  • Knowledge of the current state of immunology and immuno-oncology

Summary

Invenra is seeking a Director of Immuno-oncology. In this critical role, you will be responsible for managing teams that are investigating the in vitro and in vivo pharmacology aspects of drug discovery programs, advancing the programs to IND, and supporting mechanisms of action and new indications of molecules in the clinic. You will collaborate closely with both internal and external scientists. In this position, you will report to the Senior Vice President of Research & Development and will actively participate in the expansion of the Immuno-Oncology team.

Summary

Invenra is seeking a highly motivated and experienced scientist with a background in Protein Modeling and/or design. This individual will contribute towards the direction of our drug pipeline from candidate identification through validation. The candidate will utilize computational methods, and work with the project teams to design antigens, build models of lead candidates, identify liabilities in target sequences, and propose mutational libraries to mitigate liabilities, improve the general developability profile, and/or tune binding kinetics. The position requires excellent scientific and problem-solving skills, with a strong focus on creative solutions to technical problems.

All levels will be considered, but special consideration will be given for those with a diverse scientific background with experience in:

  • protein expression, protein purification, and assay with a desire to test computational hypotheses in the lab; or

Summary

The Protein Analytics/Characterization team at Invenra is seeking a protein LC-MS analytical Scientist or Senior Scientist to join their expanding team. This position will report to the Director of the Analytics group and provide protein mass spectrometry support for numerous projects relating to Invenra’s protein biotherapeutics development pipeline. The primary duty of this position is to perform intact mass measurements of antibodies, antigens, and other protein constructs using LC-MS techniques. Critical is the ability to troubleshoot and reconcile mass differences between predicted and measured masses and report upon these differences to colleagues and collaborators. Other duties include LC-MS assay development (bottom-up, middle-down/up, top-down), instrumentation troubleshooting (including maintenance/repair), and providing expert-level technical support to Invenra’s numerous pre-clinical protein biotherapeutic programs. Key aspects of the job entail operating an Agilent 6545XT Q-TOF mass spectrometer and 1290 UPLC to support routine protein mass analysis and product quality monitoring. As the protein mass spectrometry team grows, this position will support development of more advanced LC-MS assays to facilitate more rigorous protein characterization (peptide mapping/sequence confirmation, analysis of glycosylation and other post-translational modifications, disulfide bond mapping, drug-antibody-ratio estimation, epitope mapping, etc.).

The position requires mastery of protein mass spectrometry by LC-MS and a knowledge of basic antibody structure and methods of analysis by LC-MS. We seek a candidate with technical mastery of protein sample preparation strategies for LC-MS applications – this is a hands-on, lab-driven position with minimal large-scale bioinformatics needs. The candidate must be familiar with operation and basic maintenance of high-end LC-MS instrumentation and must have a watchful eye to identify early-stage degradation of instrument performance. Experience using LC-MS to analyze biomolecules including mono-/multi-specific antibodies, nanobodies (VHH), cell-surface proteins, and affinity-tagged proteins is highly desired. Familiarity with Agilent’s LC-MS instrumentation and data analysis software is also highly desired. The ideal candidate will skillfully interpret LC-MS results and communicate these findings to non-experts to help make major decisions about project direction.

Responsibilities

  • Characterizing antibodies, antigens, and other engineered proteins by LC-MS. Ensuring data quality meets or exceeds quality criteria.

  • Performing as the LC-MS technical lead on project teams.

  • Employing new strategies for protein characterization featuring LC-MS techniques.

  • Establishing suitable standards and controls for new assays and writing standard operating procedures for use.

  • Preparing analytical standards, chromatographic mobile phases, and reagent stocks.

  • Performing basic/routine maintenance and instrumentation troubleshooting.

  • Analyzing and interpreting data from multiple assays, compiling and reconciling those data with LC-MS outputs, and communicating results to scientific and management stakeholders.

  • Mentoring junior scientists and managing research associates.

Knowledge, Skills and Abilities

  • Mastery of protein sample preparation for LC-MS applications. Expertise in chromatographic separations of proteins and operation of sensitive analytical instrumentation is required. Experience with protein chromatography is required.

  • Expertise in top-down (intact) protein mass spectrometry is preferred.

  • Expertise performing affinity purification/immunoprecipitation is desired.

  • Required is the ability sensibly improvise and design new experiments based upon multiple analytical readbacks without significant technical oversight from the Director of Analytics.

  • Familiarity with Agilent LC-MS instrumentation and software (MassHunter, Bioconfirm) is highly desired. Experience using Agilent 1100/1200 HPLC systems is a plus.

  • Must be capable of expertly interpreting LC-MS data and presenting results to cross-functional teams comprising various scientific disciplines.

  • Should be skilled in the area of routine maintenance of LC-MS instrumentation including ionization source/ion optics cleaning, replacing pump seals/pistons, injection valve repair, and replacement of sampling needles and in-line filters. Preference will be shown for candidates with experience with Agilent HPLC and LC-MS instrumentation.

  • Must have a solid grasp of the underlying biochemistry/chemistry governing protein modification (endogenous and exogenous) and how that relates to biologically meaningful prevalence in engineered proteins from transient transfection and stable cell line expression.

  • Must execute routine work at expected quality with minimal supervisory oversight, but wise enough to know when to ask for help to avoid preventable delays to project timelines.

  • Must be willing to mentor junior scientists, manage research associates/interns, and foster a laboratory environment comprising analytical rigor, creativity, and accountability.

  • Must adhere to standard operating procedures when required.

  • Must be reliable, have a professional attitude, and maintain positive group dynamic through effective and timely communication.

Scientist level: Ph.D.’s without post-Ph.D. work experience.  Experience and technical aptitude will be considered as a substitute for formal education for candidates with a bachelor’s or master’s degree with 7+ or 5+ years of LC-MS experience, respectively, relevant to the requirements/preferences for this position.

Senior Scientist level: Senior Scientist level will (1) have a Ph.D. in analytical chemistry, biochemistry, or the biological sciences/physical sciences (molecular and cell biology, physics, etc.) with advanced training with LC-MS, and (2) two or more years of relevant industrial or post-doctoral experience in an LC-MS laboratory.

We seek a candidate with strong technical competencies regarding operation and repair of chromatographic instrumentation and expert grasp of the fundamentals of chromatographic separations including underlying chemistry. Experience analyzing/purifying biomolecules including mono-/multi-specific antibodies, nanobodies (VHH), cell-surface proteins, and affinity-tagged proteins is highly desired. Experience using and interpreting data from multiangle light scattering instrumentation (MALS) is a plus. Responsibilities include:

  • QA/QC characterization of antibodies, antigens, and other engineered proteins by HPLC using SEC, HIC, IEX, and SMAC techniques with UV, fluorescence, and/or MALS detection. Must endure data quality meets or exceeds quality criteria.
  • Affinity purifying and/or 2nd-step polishing proteins by gravity filtration, HPLC, and/or FPLC.
  • Employing new strategies for protein characterization using chromatographic techniques.
  • Developing and/or modifying purification techniques to improve protein yield, protein purity, and sample throughput.
  • Establishing suitable standards and controls for new assays and writing standard operating procedures for use.
  • Preparing analytical standards, chromatographic mobile phases, and reagent stocks.
  • Analyzing and interpreting data from multiple assays, compiling and communicating results.
  • Serving as a representative from Analytics on project core teams.

Knowledge, Skills, and Abilities:

  • Mastery of chromatographic separations and operation of analytical instrumentation is required. Experience with protein chromatography strongly preferred.
  • Must be capable of routine maintenance of HPLC/FPLC instrumentation including replacing pump seals/pistons, injection valve repair, and replacement of sampling needles and in-line filters. Preference will be shown for candidates with experience with Agilent 1100/1200 HPLC systems and GE (Cytiva) AKTA FPLC instruments.
  • The ideal candidate should be capable of advanced HPLC/FPLC instrument troubleshooting and repair including diagnosing faulty electronic boards and performing major hardware repairs (replacing pump assemblies, replacing transport carriages, rebuilding detection cells and degassers, etc.).
  • Must have solid grasp of underlying chemistry governing chromatographic separations including the variables that can be adjusted to achieve the desired experimental outcome.
  • The ability to execute routine work at expected quality with minimal supervisory oversight is a requirement.
  • Must be willing to mentor junior scientists, research associates, and interns while fostering a laboratory environment comprising analytical rigor, creativity, and accountability.
  • Must adhere to standard operating procedures when required.

Education and Experience:

The target candidate at the Scientist level will have a Ph.D. in chemistry, biochemistry, or other biological/physical science discipline (molecular and cell biology, immunology, physics etc.) with 0-2 years of industry experience. This individual will support the Invenra antibody discovery platform by helping to transition candidate multi-specific antibodies to drug leads. The successful candidate will use their knowledge of protein biochemistry to work with the translational BioProcess group to lead and meet production needs for in vivo or cell culture experiments, develop scalable and robust purification processes for challenging lead candidates, perform the experimental work, and maintain the documentation needed to support successfully transfer purification processes to a Contract Manufacturing Organization in preparation for IND submissions.

Responsibilities:

  • Work with a multidisciplinary team to execute experiments to evaluate long term developability and manufacturability of lead candidate bi-specific antibodies.
  • Collaborate with project CORE teams to interpret data from previous antibody expression, purification, and formulation studies to plan process development studies targeted at improving expression/purification yield, proper assembly, and overall purity.
  • Manage and execute routine pipeline purifications of lead candidates needed to support in vivo studies by working closely with the Translational BioProcess team and broader Invenra team (analytics, quality, project management) to ensure that these materials meet timeline and quality specifications, and all activities are documented for traceability.
  • Keep meticulous records to ensure that stage appropriate data and metrics are collected and can support activities such as CDMO collaboration for stable cell line generation, GMP manufacturing, and IND submission.
  • Assist in writing activities such as drafting SOPs for a growing group, and preparing development workplans and reports.

Education Requirements:

BS or MS in biochemistry or related field and multiple years’ work experience in a related field. Exact level and title dependent on experience.

Specific skills:

  • Practical experience in biochemistry, protein purification, biologics manufacturing and/or process development including working with automated FPLC systems and knowledge of the various chromatography and purification methods (affinity, Ion exchange, HIC, formulation with UFDF/dialysis, aseptic technique, etc.)
  • Experience and understanding of classic protein analytics and biophysical characterization techniques including HPLC-SEC, mass spectrometry, BLI/SPR, Tm, DLS, ELISA, CE-SDS, etc.
  • Strong critical analysis and troubleshooting skills to evaluate data and make decisions in the areas of protein expression, purification, formulation and analytics.
  • Excellent organization skills to document and communicate experimental strategy, design and results to team members and collaborators.
  • A general understanding of cell culture and protein expression.
  • Experience with AKTA chromatography stations and Unicorn software, programming automation high throughput liquid handlers is a plus.

  • Bring diverse experience in biochemistry, protein purification, and characterization, as well as strong critical analysis skills to contribute to the implementation of antigen-specific analytical strategies to ensure antigen quality, and determine what should be used in selections, or immunization.
