Bispecific Discovery

Outline of the Rapid Antibody Discovery Process Invenra’s Rapid Discovery Services provide an efficient, industry-leading approach to discovering monoclonal and bispecific antibodies. With cutting-edge phage display technology and our streamlined workflow, we deliver high-quality antibody leads in record time.

Rapid Bispecific Discovery

Features:

  • Inputs: Target ID(s), recombinant proteins, and cell lines
  • Deliverables: 10 unique B-Body® Bispecific Antibody sequences
  • Timeline – 4 months from DNA sequence submission to delivery

Process:

  1. Assessment – Invenra assesses target feasibility and compatibility for the Rapid Discovery Workflow
  2. Handoff – Clients supply purified recombinant antigens and necessary cell lines, or Invenra can source materials for an additional fee
  3. Library Screening – Invenra screens >30 proprietary human phage libraries under three selection conditions, conducting 90 screens in parallel
  4. Binding Validation – Cross-reactivity testing with up to three additional species/related proteins and affinity (KD) measurements
  5. Bispecific Expression – A 12×12 B-Body Bispecific matrix in two orientations (a total of 288 total combinations) is expressed, purified, and screened
  6. Selection and Assay – Top candidates are confirmed for target binding, self-association, polyreactivity, and functionality using a high-throughput screening assay
  7. Delivery – Data is shared with the client, and the top 10 B-Body Bispecific sequences are delivered

HPLC Analysis of Expressed Antibodies

93% Purity After One Step Protein A Purification

HPLC analysis illustrates the high yield and-purity possible with B-Body® Bispecific antibody expression and one-step purification. Bispecific Ab was transiently expressed in CHO cells and purified by Protein A purification. HPLC analysis provides readouts of yield and purity as well as valuable, predictive insights for analytical development, downstream processing, and formulation optimization.

Expression Performance

High Performance Across a Range of Formats and Samples

This dataset comprises 549 protein samples (~493 unique sequences) expressed in CHO at 50–200 mL scale, spanning IgG1, One Arm, 1×1, 2×1, 2×2, and trispecific formats.

Panel A shows crude yield after capture purification (x-axis) versus crude purity (y-axis), illustrating that the service can successfully express antibodies across a broad range of molecular formats and a large range of antibody specificities.

Panel B summarizes purity distributions for crude and polished samples. In each box plot, the central line represents the median, the box edges mark the 25th and 75th percentiles, and whiskers indicate the statistical minimum and maximum values (outliers excluded). In all cases, polishing improved purity, with the lowest observed values exceeding 85%, demonstrating that the service reliably delivers high-quality material across diverse antibody architectures.
